There is a black spot on the tpo left of my screen after I fixed the screen which was not turning on

It sounds as though the replacement screen was either damaged in the installation process or defective. Can you add a picture of what the black spot looks like?

Thanks for the picture, Shaik. Rereading your question and later comment, I realize I had assumed you replaced the screen with a new one, but it appears you're using the original screen.
Anyway, I'm not sure what might have caused those spots; my best guess is that adhesive may have gotten onto the OLED itself causing those spots. If it was just isopropyl alcohol from cleaning the old adhesive, there's a possibility it could recover after the alcohol dries, but if it's glue then those spots are going to be permanent.
I'd suggest you give it some time to see if the spots eventually get smaller and disappear; if not the only fix is going to be to replace the screen. You'll have to decide if you can live with those small defects or spend the money and time to replace the display.

My screen's main cable was loose ,after I fixed it it had a spot but still the touch was working . I even tried the pixel fixing software according to google
