Same issue. I checked the cable, removed the tape on the case to prevent shorting. I checked for pinches or sharp creases, and it's all good. It's so sporadic, and unpredictable. Is it still the SSD? Is it the logic board connector? If so, why do rotational work? 12 years of rebuilding, this one has to be the most frustrating.
I guess I will try an Evo, and then search Ebay for a replacement logic board. I have just run out of reasons, aside from the full Monty - the logic board.
Is there any software to totally clear the SMC and EFI so it starts fresh with an install from the OS build? Maybe, there is corruption at that level? I'm reaching now. I also thought of connecting it via a caddy in the DVD slot, and use that Sata connection?
I'm ready to try anything. Thanks.

All I get is prohibitive sign on startup, no idea why. It flashes the Apple Logo, quickly, then back to circle. It will start if I hold down shift button, but not go into safe mode.

Successfully installed El Capitan on suspect SSDs. Tried both Kingston (never had an issue in the past), and a Crucial 240. Same results, no startup. But, does see SSD and writes system to drive.

Hi all,
Desperately trying to get an SSD to be start up device. All I get is the prohibitive sign [circle with slash].
2009 2.53, 4gig ram, 15”, 2009, 5,3. 240 SSD's tried.
240 Kingston and 240 Crucial tried.
All I get is prohibitive sign on startup, no idea why. It flashes the Apple Logo, quickly, then back to circle. It will start if I hold down shift button, but not go into safe mode.
What I have done, so far, to no avail:
Never had an issues in building about 15 of these.
Successfully installed El Capitan on suspect SSDs. Tried both Kingston (never had an issue in the past), and a Crucial 240. Same results, no startup. But, does see SSD and writes system to drive.
Tried 2 different drive cables - no difference.
Hooked SSD to USB adapter - worked fine to start up.
Performed PRAM and NVRAM resets, no help.
Changed ram, no difference.
Tried rotational drive, works perfect to build and startup. But, slowwww.
Tried to update SMC and EFI firmware. “This firmware not needed for this computer” message.
I have another drive cable, due today, but I have a bad feeling it's not going to change.
