Separate headstock & neck of Rock Band Fender Stratocaster

Open the headstock

Unscrew the 4 screws at the back of the headstock.
You can also unscrew the screw at the top of the neck.
You don't have to do this last step, but it might give you a bit more wiggle room later on.
