Does water come through the brew head with the portafilter off? If not remove and clean the brew head screen; coffee grounds can get behind it and clog it. If water does flow with the portafilter off, but not with it on, you've either got a clogged basket, coffee that is too finely ground or you're tamping too hard, or some combination of all three.
