It sounds like you may be having issues with your memory card. The memory card may be faulty/defective or you could just have the "lock" setting on the memory card prohibiting you from uploading your pictures.

If this does not resolve the issue, I would think that there is something wrong with your USB cable. It also may just be defective and needs to be replaced.
